Multi-objective fluorescent molecule design with a data-physics dual-driven generative frameworkYanheng Li, Zhichen Pu, Lijiang Yang et al.
Designing fluorescent small molecules with tailored optical and physicochemical properties requires navigating vast, underexplored chemical space while satisfying multiple objectives and constraints. Conventional generate-score-screen approaches become impractical under such realistic design specifications, owing to their low search efficiency, unreliable generalizability of machine-learning prediction, and the prohibitive cost of quantum chemical calculation. Here we present LUMOS, a data-and-physics driven framework for inverse design of fluorescent molecules. LUMOS couples generator and predictor within a shared latent representation, enabling direct specification-to-molecule design and efficient exploration. Moreover, LUMOS combines neural networks with a fast time-dependent density functional theory (TD-DFT) calculation workflow to build a suite of complementary predictors spanning different trade-offs in speed, accuracy, and generalizability, enabling reliable property prediction across diverse scenarios. Finally, LUMOS employs a property-guided diffusion model integrated with multi-objective evolutionary algorithms, enabling de novo design and molecular optimization under multiple objectives and constraints. Across comprehensive benchmarks, LUMOS consistently outperforms baseline models in terms of accuracy, generalizability and physical plausibility for fluorescence property prediction, and demonstrates superior performance in multi-objective scaffold- and fragment-level molecular optimization. Further validation using TD-DFT and molecular dynamics (MD) simulations demonstrates that LUMOS can generate valid fluorophores that meet various target specifications. Overall, these results establish LUMOS as a data-physics dual-driven framework for general fluorophore inverse design.

Accelerating Locality-Driven Integration in Quantum Chemistry with Block-Structured Matrix MultiplicationXinran Wei, Yan Pan, Fusong Ju et al.
Locality-driven integration is a pervasive computational pattern in quantum chemistry, arising whenever spatially localized basis functions interact through numerical quadrature or integral screening. The dominant matrix multiplications in these tasks exhibit dynamic, structured sparsity driven by spatial locality, posing significant challenges for both dense batched kernels and generic sparse formats on GPUs. We present KerneLDI, a GPU-oriented framework that addresses this regime by co-designing data layout, screening logic, and matrix-computation operators to realize block-structured matrix multiplication for locality-driven integration. KerneLDI reorganizes operand matrices into a unified block-filtered representation that retains only spatially relevant blocks, and executes the resulting contractions with customized dense block multipliers that adapt proven dense-matmul optimizations to retained block pairs. We develop and evaluate KerneLDI on exchange--correlation (EXC) integration in Kohn--Sham density functional theory, a representative and computationally critical instance of this pattern. Across diverse molecular systems, KerneLDI preserves numerical accuracy while delivering up to 10$\times$ speedup for EXC evaluation over a dense GPU baseline, scales favorably with increasing system size and multi-GPU parallelism, accelerates end-to-end self-consistent field calculations, and yields nearly 6$\times$ throughput improvement for ab initio molecular dynamics.

Continuous Control Reinforcement Learning: Distributed Distributional DrQ AlgorithmsZehao Zhou
Distributed Distributional DrQ is a model-free and off-policy RL algorithm for continuous control tasks based on the state and observation of the agent, which is an actor-critic method with the data-augmentation and the distributional perspective of critic value function. Aim to learn to control the agent and master some tasks in a high-dimensional continuous space. DrQ-v2 uses DDPG as the backbone and achieves out-performance in various continuous control tasks. Here Distributed Distributional DrQ uses Distributed Distributional DDPG as the backbone, and this modification aims to achieve better performance in some hard continuous control tasks through the better expression ability of distributional value function and distributed actor policies.
